DiI-HDL Uptake Assay


Add DiI-HDL (final concentration 10μg/ml) to Dulbecco's Modified Eagle's Medium (CELLGRO, #10-013-CV)/Ham's F-12 medium (CELLGRO, #10-080-CV) (1:1), 50 units/ml penicillin, 50 µg/ml streptomycin, 2 mM glutamine (Invitrogen), 10% FBS (Fetal Bovine Serum, JRH Biosciences), 0.5% (w/v) BSA


Filter media through 0.22μm filter and add to cells (8-10 mls in 10-cm plate or 0.5 ml/well in 24-well plate


Incubate cells for 2 hrs at 37oC and observe with inverted fluorescence microscope
